In biomedical research, investigators usually focus on specific ceil types or analytes bound to micro-particles that are suspended in complex samples containing numerous interfering substances. Cell separation is therefore a critical technology for both basic biomedical research and clinical practice. One of the most effective approaches is to exploit dielectrophoretic (DEP) forces to steer cells along distinct paths. DEP is the electromagnetic force a cell experiences in a non-uniform electric field, and the key to this technique is to generate such afield with a combination of signal source and high-voltage amplifier. Experimental procedure: The host PC outputs a 0-5 Vpulse train; a power amplifier boosts it to 0-1 kV, creating a potential difference between the two micro-jets that establishes a non-uniform electric field and the resulting dielectrophoretic force. Meanwhile, the sample-containing suspension is continuously infused into the microfluidic chip; cells deflected by the DEP force enter the designated channel and are separated. A laser sensor downstream verifies the sorting outcome in real time.
