Modulated LED Light Source For Fluorescence Lifetime Imaging

Product catalog summary
Overview
The document describes a modulated LED light source designed for use in Fluorescence Lifetime Imaging Microscopy (FLIM) systems operating in the frequency domain. This LED light source is suitable for various microscopy systems provided by manufacturers like Nikon, Olympus, Leica, and Zeiss.

Specifications
  • Wavelengths: Available in 448 nm, 468 nm, 488 nm, 538 nm, 630 nm, with others available upon request.
  • LED Nominal Power: 3 W typical.
  • Modulation Frequency Range: 5 – 120 MHz.
  • Control: Digital control via USB interface emulating an RS232 serial port.

Power Supply/Control Unit
  • Maximum Modulation Input: 0 dBm max.
  • RF-Amplifier Gain: 40 dB typical.
  • Output Power: Max. 10 W.
  • DC Current: 0 – 1500 mA.
  • Dimensions: 257x165x256 mm.
  • Total Weight: Approx. 6 kg.

Applications
The LED light source is used in protein interaction studies involving fluorescent proteins. It facilitates the detection of energy transfer between donor and acceptor molecules, evidenced by a decrease in fluorescence lifetime of the donor molecule. An example provided shows a decrease in the lifetime of CFP from 2.4 ns to 1.9 ns when fused with YFP, indicating energy transfer.

Conclusion
This modulated LED light source is a versatile tool for FLIM applications, offering a range of wavelengths and robust control features, making it suitable for advanced microscopy studies.

Modulated LEDs of different wavelength bands are available. The output light intensity can be varied over a wide range and the maximum excitation power available at the sample is comparable with that of other light sources. The system consists of an RF-shielded LED assembly that is fitted in the standard mercury lamp housing as provided by the microscope suppliers Nikon, Olympus, Leica and Zeiss. The LED assembly is connected via a shielded cable to the Power Supply/Control Unit (PS/CU) that provides the DC and AC power to the LED. To build a complete set-up for lifetime imaging, a compatible modulation signal generator (control of modulation frequency and amplitude), modulated intensified CCD camera and dedicated image processing software are available from Lambert Instruments.
